Abstract:Over the past years, the role of genetic events in hematopoietic malignancies has been studied extensively, and many molecular defects and their contribution to leukemogenesis could be unraveled. However, the understanding of epigenetic alterations is not as advanced, but it is currently gaining strong momentum as epigenetics are likely to play a major role in leukemogenesis. DNA methylation is an important epigenetic mechanism regulating gene transcription and occurring in both normal and malignant hematopoiesis. Whereas the balance and coordination of this regulatory process is fundamentally required in normal blood cell development, it is often perturbed in hematopoietic malignancies. The first part of this chapter discusses mechanisms and targets of aberrant DNA methylation as well as their biological significance in hematopoietic disorders. In addition, the second part provides a brief overview of commonly used techniques for site-specific and global DNA methylation analyses.
