Abstract:	Understanding the mechanisms that regulate gene expression during development is a major challenge in science. In this issue of Developmental Cell, Amano and colleagues report that expression of Sonic hedgehog (Shh) protein in the posterior mesenchyme of the mouse limb bud correlates with a long-range chromatin interaction with enhancer MFCS1 and looping of the Shh locus from its chromosome territory (CT).
