Abstract:Cyclic adenosine monophosphate (cAMP) is the second messenger synthesized by adenylate cyclase (AC) inresponse to β-adrenergic receptor (βAR) stimulation. In normal conditions, cAMP plays an important role inregulating several cardiovascular functions, including heart rate and contractility. Several abnormalities in β-adrenergic signal transduction have been identified in heart failure and they are thought to be linked to thedecrease in cardiac contractility. Different mechanisms have been considered responsible for these alterations,including desensitization/down-regulation of βARs and decreases in AC5/AC6 levels, the major cardiac ACisoforms. Recently, several experimental studies have been performed on transgenic animals to clarify the roleof the different AC isoforms in normal conditions and during the development of left ventricular dysfunction.This review describes the results of the latest experiments based on the genetic manipulation of AC isoforms,and the new possible therapeutic strategies for treating heart failure which arise from them.
